Intestinal bacteria affect immune cells
According to a new study intestinal bacteria play an important role in the development of white blood cells (leukocytes) that are associated with the immune system and the basic unit to fight infections that affect the body.
The human body is home to thousands of species of microbes, which under certain conditions can cause disease, but also have a number of benefits to our health qualities.
The highest concentration and the greatest diversity of bacterial species these sows findeth in the gastrointestinal tract and in particular in the colon.
A team from the California Institute of Technology (Caltech) in Pasadena, led by Sarkis Mazmanian, a professor of biology and biological engineering, has conducted research on this area of the human body.
Their work with mice has led to many discoveries about the importance of intestinal bacteria for health.
For example, in 2013, the team reported the relationship between microbes and autism, as well as their impact on the development of multiple sclerosis.
In a recent study published in the journal Cell Host & microbe, researchers describe their discovery that the beneficial intestinal bacteria play a key role in the development of innate immune cells - macrophages, monocytes and neutrophils - special white blood cells that provide the first line of defense against invading pathogens.
When the team comparing the number of white blood cells in these areas in mice born without intestinal bacteria - known as " non -microbial " mice and normal mice with normal colonic fluorines, they found that non - microbial mice had fewer white blood cells.
The team tested the ability of mice to combat infections by exposing them to a mono-cytogenetic bacterium Listeria.
What cells of the immune system exist?
Cells of the immune system are responsible for the maintenance of the immune reactivity of the organism against penetrating viral and bacterial pathogens in the human body, which sometimes can cause dangerous health infections. Initially, the circuit is driven by the production of pluripotent stem cells that differentiate and give rise to clones of B-and T-cell lymphocytes, neutrophils, eosinophils, basophils, monocytes, dendritic cells, mast cells and red blood cells.
Neutrophils are a special type of immune system cells, which are differentiated from myeloid stem cells. They migrate into the bloodstream at himiotaksichen signal from the body. The next stage of their operation is not present on its surface MHC molecules of the system that is the major histocompatibility complex on. Also involved in the activation of clusters of differentiation in the body - CD-cells.
T lymphocytes very well may be defined as "the heads of special operations to eliminate the invasion and destruction of pathogenic microorganisms." They are produced in the bone marrow pluripotent stem cells and then migrate to the thymus and mature there. Subsequently differentiate and acquire the specific T-cell reactivity - TCR.

B-cell lymphocytes are another important cellular arm of the immune reactivity of the organism against environmental emptied the invasion of microorganisms. They are involved in humoral immunity by synthesis of antibodies. Each mature B lymphocyte produces only one type specific antibody that is expressed on the surface, such as a B cell receptor. Antibodies react against the onset of the invasion antigens.
NK-cells are a specific cell type. They can be defined as "a SWAT team to fight pathogens." Their name comes from the abbreviation natural killers, which means that they kill directly taken into the body microorganisms without subjecting them to any attempts to phagocytic or neutralization. NK-cells are most pronounced cytotoxicity compared with all other members of the immune response. Thus limiting the spread of virus-infected cells in the human body.

In my current project at Imperial College London, I am focusing on several proteins from different human pathogens. One of these proteins forms crystals, which tend to be looped with the unique c axis of ~500Å in length and oriented in parallel with the X–ray beam. This led to a severe spot–overlapping problem, which was resolved with the aid of a kappa goniometer.
Gentaur products came to the rescue in different steps of this project.
The first issue to solve was the fact that crystals grew on layers of microcrystals mixed with protein skin, which, when disturbed with a traditional loop, would break into many pieces, enter the loop and thereby abolish diffraction. The Microloops™ allowed for precise selection of the desired crystals without perturbation of the drop, so that skin-free crystals inside the loop could be obtained.

X–ray beam and ruler are represented in red.Another problem was the reproducibility of the crystals, when crystallization trials were set–up in 96, 48 or 24 well plate formats. I decided to try the new in–situ plates and to compare them with traditional 96 well, 48 well and 24 well plates. This experiment was performed on the same day and using the same protein batch. Whilst the other plates gave either only micro–crystals (<1μm) or failed completely to produce crystals within a given crystallization condition range, the Gentaur plates gave crystals of about 30μm in size (Fig.1)!

Diffraction experiments were performed in situ and crystals diffracted approximately to 3.8Å (Fig.2) with a very low background diffraction compared to competitors.
In my opinion the greatest benefits were: crystal reproducibility, crystal–friendly plate transport and low background scattering. There are a few others benefits, which are not less important, such as the possibility of shooting the crystals with better orientations for data collection or the possibility of testing lattice disorders from data collected at RT vs. cryo data. These are all applications, which I am currently exploring at the i24 beamline at the Diamond Light Source (Oxford, UK), where complete data collection is currently achievable in situ.
We all know that obtaining crystals and manipulating crystals represent a continuous challenge; therefore I would like to thank Gentaur for producing innovative products, which assist us crystallographers not only in delivering better results but also in overcoming experimental obstacles, which otherwise could severely delay or hamper our work.

Speedy Breedy turns microbial activity into data. When Speedy Breedy is connected to a PC the experiment is visualised in near real time as a graph.
Speedy Breedy comes pre-loaded with standard protocols that are designed to search for a "significant event";, that is when there is a significant pressure change in the headspace of the culture vessel, meaning there is microbial activity inside the culture chamber.
Speedy Breedy's Windows compatible software is intuitive and easy to use. Customers can design their own protocols on the PC and download them to Speedy Breedy. Protocols enable the user to set the incubation temperature (between 5 below ambient and 44 degrees), vary the time pressure readings are taken and the speed at which the mixing paddles rotate inside the culture vessel.
Experimental data can be uploaded from Speedy Breedy's SD card to the PC for archiving and further analysis.
How Speedy Breedy Works
Speedy Breedy is a portable, sensitive, precision respirometer which detects and monitors microbial activity. Detection of microbial activity is determined as a consequence of pressure transients relating to gaseous exchanges within a closed culture vessel of 50 ml working volume, as a result of microbial respiration.
Samples are added to a culture vessel containing a suitable medium and where present, contaminating microorganisms begin to grow. Speedy Breedy has advanced mixing technology that homogenises the culture conditions for rapid growth of microbes and efficiently converts gaseous changes in the culture medium into pressure variances in the headspace, therefore allowing detection of pressure transients due to metabolic processes where microbes are present.
Continuous data collected by the Speedy Breedy is analyzed in real time to provide pressure transients. Detection algorithms process these pressure transients to alert when significant changes have taken place.
Speedy Breedy measures both positive and negative pressure meaning that monitoring can be performed on a range of microbial processes reacting to differing conditions within the culture chamber.

Significant discrepancies between FISH, IHC results for ALK testing
The findings of a recent study indicate that routine testing with both fluorescent in situ hybridization (FISH) and immunohistochemistry (IHC) may enhance the detection of ALK-positive non-small cell lung cancer (NSCLC). Accurate determination of ALK-positive tumors is necessary to identify patients with advanced NSCLC who are most likely to benefit from targeted therapy with an ALK inhibitor.
The discovery of ALK rearrangement in about 1% to 7% of NSCLCs led to the development of ALK inhibitors, such as crizotinib, which have significantly improved treatment response among people with ALK-positive NSCLC. FISH with break-apart probes is currently the only approved test for the detection of ALK positivity in NSCLC, but this technique may not be available or feasible in all institutions. IHC, which is more affordable and accessible than FISH, has been found to be reliable as a screening tool, but interpretation of its results have not been standardized. Researchers continue to explore the optimal testing process for identifying ALK-positive NSCLCs.
Parallel testing with both FISH and IHC on 3,234 NSCLCs led to the identification of 150 tumors that were ALK positive by either test. The results of FISH and IHC were discordant in 80 of these tumors. As such, the use of FISH or IHC alone would have missed approximately 25% of ALK-positive cases. The findings of the study are published in the March issue of the International Association for the Study of Lung Cancer's journal, the Journal of Thoracic Oncology (JTO).
Most discordant results were FISH-positive/IHC-negative (36 cases); FISH-negative/IHC-positive results were found in 19 cases, and FISH-noncontributive/IHC-positive results were found in 15 cases. Preliminary data from 44 evaluable patients showed that treatment with an ALK inhibitor was associated with a high response rate, with response noted among patients who had tumors with discordant results (either FISH-positive/IHC-negative or FISH-negative/IHC-positive).
"Data on crizotinib response in patients who have been diagnosed differently by FISH and IHC are still preliminary. Thus, until large-scale studies in patients under therapy with crizotinib determine which testing is the most relevant to predict responses to ALK inhibition, our data support the need to routinely perform both analyses because of the difficulty in detecting the chimeric ALK protein in NSCLC and the presence of false-negative cases for each method," says lead author Florian Cabillic, PharMD, PhD, Université de Rennes, Rennes, France.
The study also demonstrated that systematic testing of NSCLC by both FISH and IHC is feasible in routine practice. In addition, the findings indicate that performing FISH routinely on all NSCLC specimens may require an automated process at various stages of the analysis.
