Staphylococcus aureus resistance unraveled
New research findings on the mechanism and the ways in which methicillin-resistant golden staph (MRZS) becomes so difficult pathogen during the last 50 years.
MRZS unaffected by atibiotitsite, including beta-lactam antibiotic drugs such as penicillin.
Experts reveal allosteric center in the X-ray structure of penicillin binding protein 2a of MRZS. Allosteric center is a site of the protein, where its activity is regulated by its binding to another molecule.
Scientists have documented that allosteric reaction is triggered by a fragment of the cell wall at a distance of 6 nanometers and activates a number of structural changes that culminate in the opening of the active side of the closed structure, allowing catalyzing enzyme.
They also documented that the new beta- lactam antibiotic ceftaroline, which was recently approved by the Administration Food and Drug Administration in the U.S., you can contact center and allosteric trigger " allosteric opening " the active side.
Thus allowing another molecule ceftaroline to enter and activate the center . This prevents the function of the enzyme and results in the destruction of MRZS . This mechanism of action of the antibiotic is unprecedented and provides many important ideas for future work on drugs to combat methicillin-resistant Staphylococcus aureus .
Gentaur's Staphylococcus products:
Enterococcus faecium VRE; vanA, titered (1 mL)
PRODUCT DESCRIPTION: Each frozen aliquot contains 1 mL of a pure, titered culture of Enterococcus faecium. The identification of this organism was confirmed by 16S sequencing. The purity of the culture was monitored by Gram staining and by additional culturing. The titer was performed on one aliquot after freezing. The freezing medium contains 15% glycerol as a cryoprotectant. Please see the Certificate of Analysis for the specific freezing medium used.
INTENDED USE: Live, titered microorganisms can be used to determine a limit of detection (LOD), in diagnostic assay development or crossreactivity studies. When used as a control for nucleic acid tests, the same protocols as those used to amplify clinical specimens should be employed.
BIOSAFETY: Enterococcus faecium is a biosafety level 2 microorganism and must be used within Biological Safety Level 2 facility or cabinet. Please consult your institution’s regulations regarding the use of this organism.
PRECAUTIONS:
- Use Universal Precautions, this organism is potentially biohazardous.
- Repetitive freezing and thawing is not recommended (aliquot material if necessary). Titer will be altered by a single freeze-thaw.
- To avoid cross-contamination, use separate pipette tips for all reagents.
RECOMMENDED STORAGE:
Titered material should be stored at -65°C or below.
