AccuPower Taq PCR PreMix
The AccuPower Taq PCR PreMix is a convenient lyophilized PCR master mix containing Taq DNA polymerase, dNTPs, reaction buffer, tracking dye, and patented stabilizer and is aliquoted in 8-strip PCR tubes.
The premix retains its activity for over a month at room temperature and is stable for two years in -20°C freezer.
AccuPower Taq PCR PreMix is available with or without tracking dye, depending on your application. If purchased with tracking dye, reactions can be loaded on agarose gels without adding loading buffer.

p40 Antibody
The p40 antibody, catalog number A00112 is provided in concentrated form or in ready-to-use as a 2ml, 7ml, or 25ml. This antibody is very important as it is helpful in the indication of several different carcinomas and has a wide-spread use in the market. We have developed the p40 antibody as a replacement antibody for the p63 antibody. The marker is highly specific for squamous basal cells and is more specific than the p63 marker. One of the most important functions of this antibody is the ability to differentiate between adeno carcinoma and squamous cell carcinoma for lung cancer. As you are aware the difference between those carcinomas will determine what avenue of treatment will be applied. The p40 is specific for indication of the squamous cell carcinoma. We will soon be releasing our Napsin A antibody for the adeno carcinoma. These two antibodies work well in conjunction with one another and they will help the pathologist determine what method of treatment is necessary.
The p40 is also an excellent antibody in the use of prostate cancer. In the early stages of prostate cancer the basal cells inside the ducts will be stained by the p40. This will help indicate the prostatic intraepithelial neoplasia (PIN) that is considered to be a pre-malignancy as it is a carcinoma in situ of the prostatic glands. As the cancer progresses the basal cells will be seen throughout the tissue as the cells become invasive. This antibody can help determine the stage of carcinoma by indicating locations of the basal cells that are stained.
The p40 antibody is also being used with breast cancer as the antibody will stain myoepithelial cells along the ducts of the mammary glands. This antibody is excellent in determining if Atypical ductal hyperplasia (ADH) is present which can indicate a risk factor for breast cancer and serves as early detection of breast cancer. This marker will also indicate Ductal Carcinoma in Situ (DCIS) similar to the way it works on prostate cancer. This is the most common non-invasive type of breast cancer. The marker will indicate DCIS by highlighting that the cells are still within the ducts. As breast cancer progresses and becomes invasive those ducts will breakdown and the myoepithelial cells will be seen throughout the tissue and become Invasive Ductal Carcinoma (IDC).

We are wanting to explore the ability of human breast cell lines to exhibit circadian rhythm. We are also interested in influence of environmental toxicants on circadian rhythm as well as general gene regulation. I felt that the Kronos dio would be the proper equipment to perform this task as it has a CO2 hook up and can monitor 3 colors at once. Therefore, I could transfect the BASIC luciferase construct using one color and the gene of interest using a different luciferase and monitor reporter activity in real time.

Based on our proprietary site-specific TARGATT technology for fast knock-in mouse (KI) generation, Applied StemCell Inc. (ASC) recently developed a TARGATT system specifically for introducing gene(s) of interest into mammalian cell lines. The unique advantage of this system is that any gene of interest can be inserted efficiently into a defined, transcriptionally-active locus with high gene expression in a single copy fashion.
Technical Details:
TARGATT technology enables highly efficient site-specific gene integration in mammalian cells and animals1, 2. This technology uses ?C31 integrase to insert any gene of interest into a docking site, pre-engineered in an intergenic region and transcriptionally active genomic locus. Our TARGATT technology improves several aspects in the generation of transgenic cell lines and animals: (1) High integration efficiency mediated by ?C31 integrase reduces time and cost; (2) Site-specific integration at a pre-selected genomic locus eliminates position effect and ensures high level expression of the transgene; (3) Integration at intergenic region ensures that no internal genes are interrupted; (4) Single copy gene integration eliminates repeat-induced gene silencing and genomic instability; (5) Site-specific integration allows a precise comparison of the effects of the transgenes among different lines. TARGATT technology can be utilized for a variety of applications including reporter gene expression, gene knockdown, disease cell and animal models.
